摘要
目的观察大鼠哮喘模型肺组织糖皮质激素受体(GR)和β-肾上腺素能受体(β-AR)水平及平喘药物对其影响。方法用放射性配基竞争结合法测定连续激发哮喘和药物治疗后各时点肺组织GR和β-AR。结果激发后第1天肺组织胞浆GR最大结合容量(Bmax)显著升高,3天后迅速降至低于正常水平,平衡解离常数(kD)值也显著降低。肺组织胞膜β-AR、Bmax和kD值激发后逐日下降,第14天明显低于对照组。地塞米松可下调GR,雾化吸入作用弱于腹腔注射,而显著上调β-AR,雾化吸入者更明显。美喘清和舒喘灵治疗可防止GR下调。结论肺组织GR和β-AR随哮喘发作而变化,糖皮质激素(GC)可下调GR和上调β-AR,吸入优于全身用药。
ObjectiveToobservethechangesofglucocorticoidreceptor(GR)andbeta-adrenoreceptor(β-AR)oflungtissueandtheefectsoforalprocaterol,salbutamolandintraperitoneal(i.p.)orinhaled(i.h.)dexamethasone(Dex).MethodTheGRorβ-ARweredeterminedbyusing3H-dexamethasoneand3H-Dihydroalprenololasligandrespectively,ingroupsofratasthmamodelsat1,3,7,14thdayandgroupsreceivedantiasthmaticsonthe7thdayafterdailyconsecutivechalenges.ResultTheGRBmaxofasthmagroupsincreasedsignificantlyafterthefirstchalenge,butdecreasedsincethethirddaytoalowerlevelthanthatofhealthycontrol.TheKDalsodecreasedsig-nificantlysincethethirdday.Theβ-ARBmaxofasthmagroupgradualydecreasedtoasignificantlowlevelonthe14thday.TheDexi.p.grouphadthelowestGRBmaxandtheDexi.h.grouphadthehighestβ-ARBmaxa-mongthetreated.Intheprocaterolandsalbutamolgroup,therehadnodecreaseofGRBmaxcomparedwiththatofcontrols.ConclusionsTheGRandβ-ARlevelsoflungtissueschangedafterdailychalenges.TheGCplayedadown-regulationroleonGRandanup-regulationroleonβ-ARinlungtissues.TheDexi.h.hadmoreadvantagesthanDexi.p.intermsofGRandβ-ARregulations.
