马氏钳蝎蝎毒分离组分Ⅲ体外抗癌活性的实验研究 被引量:11

摘要 蝎毒组分Ⅲ(scorpionvenomcompoocntⅢ,SVEⅢ)是从马氏钳蝎粗毒分离纯化获得的有效成分之一。本文用SVCⅢ处理体外培养的人喉癌HEp ̄(-2)细胞系和人直肠腺癌C1184细胞系,研究其抗癌活性。结果表明,以丝裂霉素做为阳性对照,SVCⅢ(1.6~8.0μg/ml)对两种癌细胞呈毒性和杀伤作用(P<0.01)。当SVCⅢ浓度为4.8、6.4和8.0μg/ml时,HEp ̄(-2)细胞和C1184细胞的克隆形成率降低(P<0.01),具有剂量依赖性,半数抑制浓度分别为5.01μg/ml和6.6μg/ml。较大剂量SVCⅢ处理癌细胞24h,生长抑制率呈明显增高,以后趋于坪式。SVCⅢ还使有丝分裂指数和多核率减少。分化程度较高的HEp-2细胞对SVCⅢ更为敏感。提示SVCⅢ是存在于蝎毒中的天然、高效低毒的抗癌成分。 Scorpion venom componentⅢ (SVC Ⅲ) was an effective component isolated andpurified from the scorpion (Buthus Martensii Karsch) venom crude (SVC). The antineoplastic ef-fect of SVC Ⅲ against in vitro cultured HEp ̄(-2) human laryngeal cancinoma cell and C1184rectal adenocarcinoma cell was studied. The result showed that SVC Ⅲ at a coneentititionranging from 1.6μg/ml to 8.0μg/ml possessed the cytotoxicity and killing effect on both celllines(P<0.01). When the dosages of SVC Ⅲ were 4.8μg/ml, 6.4μg/ml and 8.0μg /ml, theircolony formation rates were lower than the blank control(P<0.01),which showed the halfinhibition effect of SVC Ⅲ in a dose-dependent manner. The half inhibition concerntrations(IC(5c))onHEp ̄(-2) cell and C1184 cell were 5.e1μg/ml and 6.6lμg/ml, respanvely. The cell growth inhibitionrate was obviously increased when both cell lines were exposed to a higher dosage of SVC Ⅲfor24 hours. If cancinonia cells werecontinually treated with SVC Ⅲ, its inhibition trended to a pla-teau gradually. The reduction of the mitotic index and the mult-neclearrate was also induced bySVCⅢ. HEp ̄(-2) cell, a highly differentiated cancinoma cell, wasevenmore sensitive to SVC Ⅲ .These results indicated that SVC Ⅲ may be a natural antineoplastic agent with higher effect and lower toxicity than SVC.
