The influence of IgA-antisperm antibody on the sperm acrosome reaction

摘要 目的探讨精浆中IGA抗精子抗体对人精子顶体反应的影响。方法利用免疫珠法(IBT)筛选出 IGA抗精子抗体阳性精浆标本同正常人精子孵育,以孕酮诱发精子顶体反应;以特异性荧光标记物-络合异硫氰酸荧光素的花生凝集素(FITC-PNA)标记精子顶体,通过流式细胞仪检测精子顶体完整性。结果与IGA抗精子抗体阳性精浆孵育的精子,其孕酮诱发的顶体反应发生率明显低于正常精浆及精子培养液组(P<0.01),正常精浆组及精子培养液组间无显著性差异(P>0.05);IGA抗精子抗体阳性精浆组、正常精浆组、精子培养液组自发顶体反应的发生率无显著性差异(P>0.05)。结论免疫性不育患者精浆中的IGA抗精子抗体可以明显抑制孕酮诱发的顶体反应的发生,可能是导致不育的原因之一。 Objective To study the effect of IgA-antisperm antibody in seminal plasma on the sperm acrosome reaction (AR). Methods IBT tests were performed in present study to screen the IgA-antisperm antibody positive semen samples. The specimens then were centrifuged and the seminal plasma was isolated, spermatozoa from ten healthy donors were incubated with these samples. The FITC-PNA was used to stain the sperm acrosome.The spontaneous acrosome reaction (SAR) and the induced acrosome reaction (IAR) by progesterone were determined by means of a flow cytometer. Results The spermatozoa treated with IgA-antisperm antibody positive seminal plasma showed significantly lower IAR responses than the two control groups (P 〈0.01), but there was no significant difference between control groups in IAR (P 〉0.05) and between the three groups in SAR(P 〉0.05).Conclusion IgA-antisperm antibody in seminal plasma can inhibit progesterone induced acrosome reaction,which may result in infertility.
