MMP10、FHIT基因在食管癌中的表达及临床意义 被引量:3

摘要 目的:探讨基质金属蛋白酶10(MMP10)、脆性组氨酸三联体(FHIT)基因在食管癌中的异常表达及二者对其发生、发展的作用。方法:应用免疫组化法对54例食管癌组织及癌旁正常组织MMP10、FHIT基因的表达水平进行检测,并研究与病史及临床病理指标的关系。结果:MMP10在食管癌组织阳性表达率为35.19%(19/54),而食管癌旁正常组织为阴性表达。FHIT基因在食管癌组织中缺失率为72.22%(39/54),明显高于食管癌旁正常组织中缺失率12.96%(7/54),二者差异有统计学意义(P<0.01)。MMP10在食管癌中的表达与食管癌组织分化程度呈负相关(r=-0.697),与P-TNM分期呈正相关(r=0.85),淋巴结转移组明显高表达(P>0.05)。FHIT基因缺失与食管癌患者的性别、病史长短、吸烟、饮酒、P-TNM分期及淋巴结转移无明显关系(P>0.05)。而与食管癌组织分化程度有关(P<0.05)。结论:MMP10在食管癌组织中有较高的阳性表达率,而FHIT基因存在较高的缺失率,且二者与食管癌临床病理指标有关,提示MMP10、FHIT基因在食管癌的发生、发展中起重要作用。 Objective:To investigate the abnormal expression and alteration of MMP10 and FHIT gene on the carcinogenesis and development of esophageal cancer. Methods: Abnormalities of MMP10 and FHIT genes were mornitored in 54 esophogeal cancer specimens and adjacent normal squamous mucosa specimens of esophageal cancer by immunohistochemical method, and their correlation with illness history, pathological features was investigated. Results:The abnormal expression of MMP10 was 35.19%(19/54) in esophageal cancer tissues and no expression in precarcinomatous lesions. MMP10 expression in cancer tissue was significantly associated with stage, cell differentiation and lymph node metastasis (P〈0.05). The FHIT gene deletion in esophageal cancer tissues was 72.22%(39/54), whereas it was 12.96% (7/54) in adjacent normal tissues, suggesting significant difference between the two groups (P〈0.01). There was no significant correlation between the deletion of FHIT gene and sex, illness time, smoking history, P-TNM staging and lymph node metastasis (P〉0.05), whereas there was significant difference between the deletion of FHIT gene and tumor cell differentiation (P〈0.05). Conclusion: The abnormal expression of MMP10 and FHIT gene is involved in tumorigenesis and development of esophageal cancer.
