热休克蛋白70表达对未成熟心肌保护作用的实验研究 被引量:5

摘要 目的探讨热休克蛋白70(HSP70)在未成熟心肌中的表达规律和对未成熟心肌的影响。方法健康新生长耳大白兔30只随机分为5组。对照组:腹腔注射生理盐水0.4 ml 24h后取离体心脏, 建立Langendorff离体心脏灌注模型;E4h、E12h、E24h、E48h组,腹腔注射去甲肾上腺素,4、12、24、48 h后分别取离体心脏,方法同对照组。测定心肌细胞中HSP70含量、血流动力学指标、生化指标,并观察心肌超微结构。结果 HSP70含量E24h组明显高于其他各组,血流动力学、生化指标均优于其他各组(P<0.01),心肌超微结构损伤较其他各组明显减轻。结论腹腔注射去甲肾上腺素24 h后可诱导未成熟心肌HSP70高表达,一定量的HSP70表达可明显减轻未成熟心肌缺血再灌注损伤。 Objective To investigate the protective effects of heat shock protein 70 ( HSP70 ) on immature myocardium. Methotis 30 rabbits (aged 14 - 21 days) were randomly divided into 5 groups. Control group (C), isolated working rabbit heart model 24 hours after intraperitoneal injection of distilled water. E4h group, E12h group, E24h group and E48h group : isolated working rabbit heart model after 4 h, 12 h, 24 h, 48 h intreaperitoneal infections of norepinephrine, respectively. Tne HSP70 content, the recovery of hemodynamics, biochemical index and uhrastrocture were studied. Results HSP70 content in E24h group is higher than other groups. The recovery of hemodynamics and biochemical index in E24h group is higher than other groups. The myocardial ultrastructure injury in E24h group is less than other groups. Conclusion This study demonstrated that HSP70 has obviously protective effects for immature mvocardium.
