摘要
目的探讨参附注射液对兔离体肺缺血/再灌注保护作用及可能的作用机制。方法将16只健康家兔随机分为两组,对照组肺以改良低钾右旋糖酐(LPD)液灌注及保存,实验组则将参附注射液(40 mg/L)加入改良LPD液灌注及保存,在4℃保存12 h后再灌注1 h,测定动脉血氧分压(PaO2)和肺气道峰压(PawP)及肺组织的湿干比(W/D);肺组织中髓过氧化物酶(MPO)活性、NF-κB的表达并观察肺组织病理结构的变化。结果再灌注15 min后,两组的PaO2逐渐降低、PawP逐渐升高,但实验组PaO2的降低程度和PawP的升高程度明显低于对照组(P<0.01);再灌注后,实验组的W/D和MPO及肺组织中NF-κB的表达也明显低于对照组(P<0.01),肺组织病理变化较对照组轻。结论参附注射液能减轻肺缺血/再灌注损伤,改善肺功能,对供肺具有保护作用。
Objective To investigate the protective effects of shen-fu injection on rabbit lung during ischemia-reperfusion, and explore its possible mechanism. Methods Sixteen rabbits were randomly divided into two groups: the control group and the trail group. The harvested lung bloc.ks were flushed with and stored in the modified low- postasssium-dextron(LPD) solution in control group, but in the trail group LPD containing shen-fu injection(40 mg/L) was used. The lung were stored at 4°C for 12 h in both groups. The isolated rabbit lung reperfusion models were established and the lungs were perfused for 1 h. PaO2 and PawP were measured at every 15 min intervals during repeffusion. After reperfusion, the lung tissue wet-to-dry(W/D) ratio and myeloperoxidase(MPO) activity were obtained. The protein expression of nuclear factor-kappa B(NF-κB) was also detected by using immunohistochemistry at the end of reperfusion. Light microscopic and ultrastructural changes were also examined. Results The level of decreased PaO2 and increased PawP in trail group were lower than in control group at the every interval time in the samples obtained 15 min after reperfusion(P〈0.01). After reperfusion the W/D, MPO, and the protein expression of NF-κB were decreased evidently in trail group (P〈0.01). The morphologic and ultrastructual damages in trail group were milder than in control group. Condusion Shen-fu injection can alleviate the ischemia-reperfusion injury of lung and improve the protective effect of lung preservation.
