摘要
以曼地亚红豆杉枝叶醇提物为材料,先经反相层析粗分离,再用氧化水解法处理含糖基紫杉烷洗脱部位,正交试验优化反应条件。结果表明,利用高碘酸钠或铋酸钠对木糖甙组分的选择性氧化,控制pH值为3左右,30℃下反应36 h,紫杉醇的表观回收率达到163%,另一个类似物10-去乙酰紫杉醇达到142%,混合组分中半合成前体巴卡亭III的含量并未因此处理而明显损失。
Selective oxidation-hydrolysis on the cleavage of the xylosyl taxanes was conducted to enhance taxol yields in Taxus media methanol extracts. The result indicated that the yields of 163 % for taxol, 142% for 10-deacetyl-taxol were achieved with the optimum reaction conditions, in which NaBiO3 or NaIO4 was used as oxidizing agent, at 30℃ with a pH of 3 within 30 hours. Moreover, the losses of baccatin Ⅲ in experimental material was not observed in this process.
