摘要
目的研究多重耐药铜绿假单胞菌PA1278所产超广谱β-内酰胺酶(ESBLs)基因序列,并确定其所产ESBLs亚型。方法琼脂稀释法进行抗菌药物敏感试验,PCR法扩增GES型ESBLs基因,并对PCR产物进行序列分析,确定亚型。结果该菌株除对碳青酶烯类中介外,对多种抗菌药物耐药,PCR扩增GES基因阳性,基因片段长543个核苷酸,核苷酸及相应的氨基酸序列经BLASTn程序比对无相同序列。结论铜绿假单胞菌PA1278所产GES型ESBLs为一种新发现的GES亚型基因(GES-1 like)。
OBJECTIVE To identify the genotype of the extended spectrum β-lactamases (ESBLs) produced by a multi-resistant Pseudomonas aeruginosa (PA1278), METHODS Antimicrobial susceptibility tests were performed by the microdilution method and the encoding gene of ESBLs was amplified by PCR. The purified PCR products were sequenced by ABI automated sequencer. RESULTS PA1278 were resistant to all β-1actams antibiotics, only sensitive to carbapenems, such as to imipenem and meropenem. The encoding genes of ESBLs were amplified as GES by PCR and the product had 543 nucleotides. Its corresponding amino acid sequence was submitted to BLAST 2.0 and found no similarity. CONCLUSIONS The β-lactamases produced by the PA1278 are novel GES-1 like ESBLs (GenBank accession number; AY953375).
