摘要
应用荧光光谱法测定人体血浆中异丙酚浓度以及一次静脉给药后人体内药代动力学参数。在含不同量异丙酚的人体血浆中加入磷酸缓冲液及环已烷/正丁醇,振摇、离心后取上清液测荧光强度(发射波310nm,激发波265nm)。结果表明异丙酚投入量(0.05~15μg)与测得的荧光强度间有良好的线性关系(r=0.9997)。提示异丙酚在人体代谢过程中符合药代动学三室模型。该方法灵敏度高,线性范围大,操作简便,专一性好,可用于临床异丙酚血药浓度监测。
the plasma level of propofol was determined spectrofluorometrically in surgical patients,which propofol had been administered intravenously,The fluorescent intensities obained from dipotassium hydrogen phosphate,cyclohexane/normal-butanol mixed to the plansma (Excitation and emission peak wavelength were at 265nm and 310 nm respectively).The experiemtal data exhibited a good linear relationship between the fluorescent intensites and the amount of propofol added to the blank plasma in the scale of 0.05~15μg,r=0.9997).The pharmacokinetic characteristic of propofol was found to the three-compartment open model.The method showed that it was quick,simple,sensitive and precise. It is suitable for clinic purpose.
