摘要
运用mRNA差异显示技术,对水稻滇一型雄性不育系和保持系花药基因表达差异进行了比较研究。以专门设计用于差异显示的90对PCR引物组合,分析滇一型10对不同品系的不育系和保持系花药基因表达差异。结果表明:这90个引物组合均有扩增产物,扩增的cDNA片段在100—2000bp之间;不育系和保持系花药cDNA扩增带型相似。在这90个引物组合中仅有15对引物组合在单对的不育系和保持系间显示多态性,而且大多数的多态性引物组合仅扩增出一条差异带。在所找到的差异中,一般总是保持系有一条带,而不育系没有对应的带或是一条很弱的带。该结果表明不育系和保持系花药mRNA差异很少。所幸的是,在这些引物组合中,只有T8×P1引物组合(5'-ATGCTGAGTGATATCTITITITITG×5'-ATTAACCCTCACTAAATGCTGGGGA-3’)在10对不育系和保持系问扩增出一个共同的差异cDNA片断:保持系在750~1000bp之间有一条带,而不育系没有对应的带或是一条表达很弱的带,这很可能就是保持系和不育系的育性基因表达差异的结果。
Gene expression in anthers of rice cytoplasmic male sterile (CMS) lines of Dian-type-1 ( D1 ) was studied on 10 pairs of CMS-D1 lines and maintainers with mRNA differential display, in which a set of RT-PCR primer combinations from Delt Differential Display Kit were screened. Within 90 combinations of PCR primers designed for differential display, only 15 primer combinations showed polymorphism between a few of the CMS lines and maintainers. Among the polymorphic primer combinations, one primer combination, T8 × P1, 5'-CATTATGCTGAGTGATATCTTTTTITITGC-3' × 5'-ATTAACCCTCAC- TAAATGCTGGGGA-3') revealed the same difference between CMS lines and maintainers of 10 pairs. Every maintainer showed a cDNA band with molecular weight between 750 bp and 1 000 bp amplified by the primer combination, while CMS lines did not have the corresponding ones or only had a very weak one. The cDNA could be related to the gene conferring cytoplasmic male sterility.
