摘要
目的探讨全反式维甲酸对肾间质纤维化大鼠肾间质结缔组织生长因子(CTGF)的影响及其可能机制。方法采用单侧输尿管结扎(UUO)大鼠模型。将40只SD大鼠随机分为5组:假手术组(A组)、UUO组(B组)、UUO+苯那普利组(C组)、UUO+维甲酸小剂量组(D组)、UUO+维甲酸大剂量组(E组)。于造模前1d分别给C、D、E组苯那普利10mg/(kg·d)、全反式维甲酸10mg/(kg·d)和20mg/(kg·d)。A、B组给予同等剂量的生理盐水。于术后第14d取术侧肾组织行HE、Masson染色评定各组肾小管间质损害程度,免疫组织化学半定量检测各组肾间质CTGF、转化生长因子-β1(TGF-β1)、α-平滑肌肌动蛋白(α-SMA)及Ⅲ型胶原(colⅢ)的表达。结果各造模组CTGF、TGF-β1、α-SMA及colⅢ的表达及肾小管间质损伤指数明显高于A组(P<0.05),而全反式维甲酸或苯那普利均能明显抑制此变化。结论维甲酸及苯那普利可能通过下调CTGF而减轻肾间质纤维化。
Objective To investigate the effect and mechanisms of all-trans retinoic acid (at-RA) on expression of connective tissue growth factor(CTGF) in rats with renal tubulointerstitial fibrosis. MethodsThe animal model of unilateral ureteral obstructive(UUO) in rats were used for the study. Forty SD rats were randomly assigned into 5 groups : sham group(group &),UUO group (group B),UUO+ benazepril treating group (group C)and two UUO+ atRA treating groups (group D,E).From the day before operation, the rats underuent intragastric administration of benazepril 10mg/(kg·d) in C group, and by at-RA 10mg/(kg· d) in group D and 20mg/(kg· d) in group E and same doses of sodium chloride in group A and B. The obstructed kidneys were sampled on day 14 after surgery. The degree of tubulointerstitial damage was scored by HE and Masson staining. The expression of CTGF, tumor growth factor- β (TGF- β ),α -smooth mused actin (α-SMA) and collagen Ⅲ (col Ⅲ) was assessed by immunohistoehemistry staining, Results The levels of CTGF, TGF-β1, α-SMA and col Ⅲ in the UUO groups significantly increased compared to those in group A(P〈0. 05) and paralled with the degree of tubulointerstitial damage, at-RA and benazepril inhibited the changes, Conclusion Reduction of interstitial CTGF expression may be an important event by which at RA prevents renal injury caused by UUO.
