摘要 目的:建立血浆中腺苷(adenosine,AD)浓度的HPLC测定方法,辅助某些药物作用机制的研究及疗效的评价,并考察腺苷在血浆中的稳定性。方法:采用反相高效液相色谱-紫外检测法测定家免血浆腺苷含量,色谱柱为YWG-C18柱(10μm,250mm×4.6mm),流动相为磷酸盐缓冲液-甲醇(80:20,pH5.9),流速为1mL·min^-1,检测波长:254nm,柱温:35℃。结果:腺苷的线性范围为0.50~40.00mg·L^-1(r=0.998(1),检测限为0.50mg·L^-1。结论:腺苷可被血液成分摄取和灭活,应用本实验建立的采样方法以及立即进行预处理可以显著减少这一过程。保证了结果的可靠性,而且检测方法灵敏度高,特异性强,操作简单,测定周期短,以此为契机推广到人体,可以为辅助药物作用机制的研究提供一种优良方法。 OBJECTIVE To determine the concentration of adenosine in plasma by RP HPLC with an attempt to study the stability,and also providing an aid for exploring the mechanism of some clinical drugs and evaluating therapeutic efficiency. METHODS An isocratic elution of a phosphate buffer and methanol (80:20, pH 5. 9) was used as mobile phase at a flow rate of 1 mL·min^-1. Detection was carried out on YWG-C18 column(10 μm,250 mm×4. 6 mm)at 35℃ with a UV detector monitored at 254 nm. RESULTS The curve was linear over the range of 0. 50 - 40. 00mg · L^-1 (r = 0. 998 0). The limits of detection (LOD) was 0. 50 mg · L^-1. CONCLUSION The application of sampling and pretreatment established in this study could decrease the inactivation of adenosine by components in blood. The assay in this study is sensitive, specific, simple and rapid. It could be a Rood assistant method for the purpose of exploring the mechanism of some therapeutic drugs.
