食管鳞癌及癌旁粘膜中核转录因子κBp65及IκBα基因和蛋白表达及意义 被引量:1

The Expression and Significance of p65 Gene and Protein of Nuclear Transcription Factor Kappa B (NF-κB) and IκBα Gene and Protein in Esophageal Squamous Cell Carcinoma and Paraneoplastic Mucosa

摘要 目的:探讨核转录因子κB(NF-κB)p65基因及其抑制基因IκBα的表达与食管鳞癌发生的关系。方法:用RT-PCR及免疫组化SP法检测了61例食管鳞癌及其正常粘膜上皮、癌旁粘膜上皮中NF-κBp65基因、IκBα基因mRNA及蛋白的表达。结果:食管鳞癌中p65mRNA、IκBαmRNA的表达量明显高于正常上皮(P<0.01),p65蛋白、IκBα蛋白胞浆表达或胞核表达率均明显高于正常上皮(P<0.01)。癌旁粘膜中,NF-κBp65蛋白、IκBα蛋白在癌旁正常上皮中的胞浆或胞核阳性率均明显低于不典型增生上皮、原位癌和浸润癌(P均<0.05)。结论:NF-κBp65、IκBαmRNA表达和蛋白表达均与食管鳞癌的发生有关。 Objective: To study the relationship between the expression of p65gene of nuclear transcription factor kappa B (NF-κB) and its suppressor gene-IκBα and in the occurrence of esophageal squamous cell carcinoma. Methods: The expressions of p65 mRNA of NF-κB and IκBα mRNA were detected using RT-PCR and the p65 protein of NF-κB and IκBα protein were detected by S-P imm unohistochemical method in specimens of 61 cases of esophageal squamous cell carcinoma and the normal and paraneoplastic epithelial mocosa. Results: The positive counts (the brightness value of band) of the expression of p65 mRNA and IκBα mRNA were significantly higher in cancer tissues than that in normal epithelium tissues (P〈0.01). The positive rates of p65 protein and IκBα protein in plasm or nuclear in cancer tissues was significantly higher than that in normal epithelium tissues yet (P〈0.01).In the tumor-adjacent mucosa,the positive rates of p65 protein and IκBα protein expression in plasma or nucleus in the normal esophageal epithelium was significant lower than that in mild and severe atypical hyperplastic epithelium, carcinoma in situ and infiltrated carcinoma. There was a significant difference between the normal epithelium and the mild and severe atypical hyperplastic epithelium, carcinoma in situ and infiltrated carcinoma (P〈0.05). Conclusion: The expressions Of p65 mRNA and protein of NF-κB, IκBα mRNA and protein may be correlated with the occurrence of esophageal squamous cell carcinoma.
