凋亡和相关蛋白bcl-2在肺癌组织中的表达及对预后的影响 被引量:10

摘要 目的:研究细胞凋亡及凋亡相关蛋白bcl-2在肺癌组织中的表达水平,及其对预后的影响。方法:应用DNA缺口末端标记技术和免疫组化方法检测116例肺癌患者组织中细胞凋亡和bcl-2蛋白表达水平。结果:116例肺癌患者中,细胞凋亡数量达到或超过0·1%的病例占48·3%(56/116),bcl-2蛋白阳性表达的病例占51·7%(60/116);单因素分析显示,肺癌患者预后的影响因素有凋亡(χ2=8·31,P=0·0039)、bcl-2(χ2=4·41,P=0·0357)、淋巴结转移(χ2=19·34,P=0·0001)和TNM分期(χ2=18·54,P=0·0001);COX模型多因素分析显示,淋巴结转移(χ2=19·28,P=0·0010)和bcl-2蛋白阳性表达(χ2=5·80,P=0·0160)是肺癌患者的预后不良因素。结论:凋亡及凋亡相关蛋白bcl-2影响肺癌患者的生物学行为及预后。 OBJECTIVE:To investigate prognostic significance of the expressions of apoptosis and its correlating protein bcl-2 in lung cancer. METHODS: The expression of apoptosis and the positive expression of bcl-2 protein were observed in 116 cases of lung cancer by using DNA nick end labeling (TUNEL) technique and immunohistochemical staining. RESULTS: In the specimens of 116 lung cancer,the expressions of cell apoptosis equal to or more than 0.1% were 48. 28%(56/116), and the positive rates of bcl-2 protein expressions was 51.72% (60/116). Single factor analysis showed that apoptosis (x^2 = 8. 31, P = 0. 003 9), bcl-2 (x^2 = 4.41, P=0.035 7), the metastasis of lymph node (x^2=19.34,P= 0.000 1) and TNM stage (x^2=18.54,P=0.000 1) affected the prognosis of lung cancer patients. COX model showed that the metastasis of lymph node (x^2= 19.28,P=0. 001 0) and the positive rate of bcl-2 protein (x^2 =5.80,P=0. 016 0) were poor factors to the prognosis of lung cancer patients. CONCLUSIONS: Apoptosis and the correlated protein bcl-2 are the factors that affect the biology behavior and prognosis of lung cancer patients.
