普伐他汀对激素性坏死股骨头内Cbfa1表达的影响 被引量:8

The Effect of Pravastatin on Expression of Core Binding Factor Alphal 1 of Steroid Induced Necrosis of Femoral Head

摘要 目的研究普伐他汀对激素性坏死股骨头内Cbfa1基因表达的影响.方法54只新西兰白兔随机分为正常组(A组)、模型组(B组)和普伐他汀治疗组(C组),每组18只.B、C两组应用大肠杆菌内毒素及甲强龙造成股骨头坏死模型,C组于造模后4周开始用普伐他汀灌胃,A、B两组以等量蒸馏水灌胃.分别于喂药后第4、8、12周分批取兔股骨头行实时荧光定量PCR检测股骨头内Cbfa1 mRNA的表达.结果B、C两组各时相点Cbfa1 mRNA的表达水平均低于A组,但C组喂药后第8、12周的表达量明显高于B组,差异有显著性(P(0.01).结论普伐他汀可促进激素性坏死股骨头内Cbfa1 mRNA的表达,有可能成为临床治疗激素性股骨头坏死的有效药物. Objective To investigate the effect of pravastatin on expression of core binding factor alphal 1 of steroid induced necrosis of femoral head. Methods 54 New Zealand white rabbits were randomly and equally divided into a normal control group (Group A), a model group (Group B) and a treatment group (Group C). All rabbits in Group B and Group C were injected lipopolysaccharide and methylprednisolone. Animals in Group C were treated with pravastatin at the 5th week, and the same volume of distilled water were given in Group A and Group B. Six rabbits in each group were killed at the 8th, 12th and 16th week respectively. Femoral head specimens were studied by quantitative real - time PCR for detecting the expression of Cbfal mRNA. Results In Group B and Group C, Cbfal mRNA expression levels were lower than that of Group A, but the expression level of Group C increased significantly compared with Group B at the 12th and 16th week ( P 〈0. Ol ) . Conclusion Pravastatin can improve the expression of Cbfal, which is probably an effective clinical treatment of early steroid -induced necrosis of femoral head.
