摘要
目的:探讨了血清IL-8和中性粒细胞趋化指数测定在脑缺血再灌注损伤中的变化。方法:采用改良Zealonga线栓法大鼠大脑中动脉闭塞(middlecerebralarteryocclusion,MCAO)模型,将大脑中动脉(MCA)先行闭塞2h,然后进行灌注不同的时间。应用免疫法测定实验组和对照组血浆中性粒细胞趋化指数,ELISA法测定IL-8。结果:血浆中性粒细胞趋化指数和IL-8于再灌注1h最高,随后缓慢下降,再灌注48h降至正常组水平。结论:中性粒细胞参与了脑组织的正常代谢及生理功能,又参与了脑缺血再灌注损伤的病理生理过程。
Objective To study the changes of neutrophil chemotactic index and serum IL - 8 contents after cerebral ischemia reperfusion injury in rats. Methods Sixty rat middle cerebral artery occlusion (MCAO) models were established with modified zealonga method. The MCA was occluded for 2 hours and reperfused for one hour ( n = 30) and 48hrs ( n = 30). The neutrophil chemotactic index was examined and serum IL-8 levels detected with ELISA (also in 30 normal rats as controls). Results The neutrophil chemotactic index and serum IL -8 levels reached the peak at 1 hour after reperfusion (P 〈 0.01 ), and then decreased. It returned to the values of the control group at 48 hours after reperfusion. Conclusion The increased serum IL - 8 levels and neutrophil chemotactic index might contribute to the development of low - perfusion reperfusion injury after experimental cerebral ischemic events in rat models.
