葛根素抗氧化作用与改善胰岛素抵抗综合征模型大鼠胰岛素敏感性、血压和血脂作用的实验研究 被引量:15

摘要 目的观察葛根素的抗氧化作用与改善胰岛素抵抗综合征(insulin-resistantsyndromeIRS)模型大鼠胰岛素敏感性、血压和血脂之间的关系。方法采用高脂、高糖和高盐饲料喂养大鼠,饲养8周时,肌肉注射葛根素,每天1次,连续4周。于造模前、造模8周后和给药4周末测定大鼠血压,并于给药4周末测量大鼠血脂、丙二醛(MDA)、超氧化物岐化酶(SOD)活性,空腹血清血糖(FBG)、空腹血清胰岛素(FINS)、计算胰岛素敏感指数(ISI)。结果葛根素高、中剂量组均能明显降低IRS模型大鼠血压和血脂;高剂量组显著提高胰岛素敏感指数和SOD活性,降低MDA水平。结论葛根素对IRS模型大鼠具有提高胰岛素敏感性、降低血压和调节血脂作用,其作用机制可能与抗氧化作用有关。 Objective To observe the effects of puerarin on antioxidation and improvements of insulin sensitivity,blood pressure and serum lipid in model rats of insulin resistant syndrome. Method adult SD rats were fed on high-fat-sugar-salt diet for 12th weeks,and puerarin was administered to the rats for 4- weeks by intramuscular injection at the begining of 9th week. BP was measured in the end of 0,8th,12th week. The levels of serum glucose,insulin,lipid,malondialdehyde (MDA) and the activity of superoxide dismutase(SOD) were measured,and the inanlin-sensitivity index was calculated at the end of the experiment.Result Experimental results indicate that large dosage of puerarin can significantly improve the insulin sensitivity and the activity of superoxide dismutase, decrease malondialdehyde. Conclusion large and/or middle dosage of puerarin can significantly drcrease blood pressure and the level of serum lipid. These effects may be concerned with antioxidation produced by Puerarin.
