周期性牵张诱导逼尿肌细胞高表达Cx43的实验研究

摘要 目的探讨周期性牵张对膀胱逼尿肌细胞表达连接蛋白43(Connexin43,Cx43)的诱导作用。方法运用牵张膀胱逼尿肌细胞体外模型,给予逼尿肌细胞周期性20%拉伸度的牵张,持续6 h;采用W estern b lotting、RT-PCR在蛋白和mRNA水平测定逼尿肌细胞Cx43表达。结果正常对照组Cx43表达量为(1 1.377 5±0.425 1),Cx43mRNA相对表达量为(0.423 3±0.187 5);而给予20%牵张刺激6 h逼尿肌细胞Cx43表达量增加至(14.015 0±0.526 2),Cx43mRNA相对表达量增加到(0.935 0±0.020 67),二者均较对照组有显著增加(P<0.01)。结论周期性牵张可显著诱导增加逼尿肌细胞Cx43在蛋白和基因水平的表达。 Objective To investigate the connexin43 overexpression of detrusor smooth muscle cells (DSMCs) by cyclic stretch. Methods The DSMCs growing on collagen-coated silicone membrane were subjected to cyclic stretch-relaxation. The Cx43 and Cx43 mRNA expression in DSMCs were detected with Western blotting and RT-PCR. Results The Cx43 and Cx43 mRNA expression of control DSMCs were respectively (11. 377 5 ±0.425 1 ) and (0.423 ± 3 0. 187 5 ), and that of DSMCs subjected to cyclic stretch-relaxation were (14.015 0 ± 0.526 2) and (0.935 0 ± 0.020 67 ), with significant difference between the two groups (P 〈0.01 ). Conclusion The connexin43 overexpression in DSMCs could be induced by cyclic stretch.
