摘要
目的:观察山莨菪碱对肝纤维化大鼠肝组织内转化生长因子-β1(TGF-β1)及其受体(TβRⅠ)表达的影响,探讨该药对肝纤维化防治作用的机制。方法:本研究用四氯化碳(CC l4)诱导肝纤维化模型。防治组在造模后1h始以山莨菪碱7.0mg.kg-1.d-1腹腔注射。大、小剂量治疗组造模后第4周始腹腔注射给药,剂量分别为14.0mg.kg-1.d-1和7.0mg.kg-1.d-1。第10周末测定血清血清谷丙转氨酶(ALT)和透明质酸(HA),RT-PCR法和免疫组化法分别检测肝组织中TβRⅠmRNA和TGF-β1蛋白的表达。结果:各山莨菪碱给药组大鼠血清ALT和HA含量均较模型组明显下降(P<0.01或P<0.05);同时该药还能抑制纤维化肝组织中TβRⅠmRNA的表达和TGF-β1蛋白的表达。结论:山莨菪碱能通过抑制肝组织中TGF-β1及TβRⅠ的表达,减轻肝纤维化。
Objective: To investigate the effects of anisodamine on the expression of TGF - β1 and its receptor of experimental liver fibrosis. Methods:The experimental liver fibrosis model was produced by CCl4. The preventive group was treated for ten weeks by anisodamine 7.0mg·kg^-1 intraperitoneal injection once everyday. All therapeutic groups were treated for six weeks by anisodamine 7.0mg ·kg^-1 or 14.0·kg^-1 intraperitoneal injection once everyday. Studies were made after ten weeks of CCl4 administration. We compared the model group with the experimental groups which were treated by anisodamine on the content of ALT and HA in sera. The reverse transcription polymerase chain reactions and the immunohistochemistry were used to detect the expressions of TβR Ⅰ mRNA and the protein of TGF - β1 in liver. Results:The serum of ALT and HA was significantly improved in anisodamine preventive group and anisodamine therapeutic groups. The expressions of TβR Ⅰ mRNA and the protein of TGF- β1 in fibrosis livers were inhibited by treatment with anisodamine. Conclusion:Anisodamine can reduce the expressions of TβR Ⅰ mRNA and the protein of TGF -β1 to prevent liver fibrosis.
