摘要
目的:建立柱前衍生化HPLC法测定复方布洛芬片中精氨酸的含量。方法:精氨酸经邻苯二甲醛和3巯基丙酸衍生化后,采用LichrospherC18(250mm×4.6mm,5μm)色谱柱,以pH7.3的磷酸盐缓冲液(取磷酸二氢钾2.0g,加水1000mL溶解后,加磷酸0.45mL,三乙胺3.0mL,摇匀,经孔径0.45μm的滤膜过滤)-四氢呋喃-乙腈-甲醇(747.5∶2.5∶75∶175)为流动相分离后以紫外330nm波长检测测定。结果:精氨酸浓度在50.0~200.0mg/L范围内,色谱响应线性良好(r=0.9997),片剂中精氨酸含量测定的平均回收率为100.1%。精密度良好(RSD=1.47%)。结论:柱前衍生化HPLC法测定复方布洛芬片剂中精氨酸的含量结果准确灵敏、方法可靠。
Objective: To establish a pre-colunm derivatization HPLC method for determination of the content of arginine in compound ibuprofen tablets. Methods: Arginine was derivatized with OPA and 3- mercaptopropionic acid. The separation was performed on a Lichrospher-C18 column (250 mm× 4.6 mm, 5 μm) with pH 7.3 phosphate buffer (2.0 g potassium dihydrogen phosphate dissolved in 1 000 mL of water and mixed with 0.45 mL phosphoric acid and 3.0 mL triethylamine) : tetrabydrofuran: acetonitrile: methanol (747.5:2.5:75: 175) mixture as mobile phase at a flow rate of 1.0 mL/min and UV detection at 330 rim. Results: The calibration curve was linear in the range of 50.0 - 200.0 mg/L ( r = 0.9997). The average recovery of arginine was ( 100.1 ± 1.47) %. Conclusions: The pre-column derivatization HPLC method was sensitive, accurate and reliable for the quantitation of arginine in compound ibuprofen tablets.
