摘要
利用PCR扩增技术筛查双价抗虫棉晋棉38的单株,对确证的转基因植株的不同生育期和不同的组织器官分别通过叶片离体饲喂棉铃虫进行抗虫性检测和通过ELISA技术检测抗虫基因的表达量。结果表明晋棉38不同发育期的不同器官中Bt杀虫蛋白含量在苗期含量较高,蕾铃期则有所下降;并且在苗期叶片的杀虫蛋白含量为最高,茎和根次之;在蕾铃期各器官的表达量顺序为:叶>苞叶>茎>花瓣>花丝>铃内。整个生长发育期的抗虫性前期抗性高于后期,抗虫性显示下降的趋势。然而抗虫棉晋棉38在整个生育期的抗虫性均为高抗水平,在各个组织器官的表达量也较高。
The positive transgenic plant of Jinmian 38 was selected by PCR detection. The Cry1Ac expression from different tissues or organs of these plants during various periods were checked through ELISA method and the leaves detached from these plants during different periods were raised in order to examine challenge to bollworms. The results suggested that the level of Cry1Ac in seedling should be higher than other periods, the expression of this protein gradually decreased from seedling to mature and the concentration of the protein was highest in seedling leaves among all tissues and organs, expression levels was gradually reduced among leave, bract, stem, petal, filament and ovule. The resistance to pest also gradually decreased from seedling to mature, but the resistance among all periods was high.
