摘要
目的研究去细胞带瓣牛颈静脉组织工程支架的制备,并观察其形态结构。方法取新鲜牛颈静脉48条,随机分为对照组、实验组,每组24条。对照组为新鲜牛颈静脉;实验组为去细胞牛颈静脉,用40 g/L脱氧胆酸钠+50 m l/L Triton-X-100为主要成分的试剂进行处理,去除内皮细胞及成纤维细胞。HE染色、苦味酸-天狼猩红染色、弹力纤维染色、电子扫描显微镜检查、基因组DNA检测,观察其形态结构,评价去细胞效果。结果实验组瓣膜及血管壁中内皮细胞、成纤维细胞去除完全,无细胞核碎片;瓣膜及血管壁胶原纤维和弹力纤维呈波浪状排列、整齐,结构完整;瓣膜、血管壁基因组DNA含量,分别较对照组下降97.58%、97.25%。结论脱氧胆酸钠+Triton-X-100是一种有效的去除牛颈静脉细胞,制备组织工程带瓣管道支架的方法。
AIM To study the decellularization procedures for bovine jugular venous valved conduits and observe their morphology and construction. METHODS Forty eight fresh bovine jugular veins were randomly divided into control and experimental group, each group had 24 veins. Control group were fresh bovine jugular veins, experimental group were decellularized bovine jugular veins. Bovine jugular veins were treated mainly with sodium deoxycholate (40 g/L) plus Triton-X-100 (50 ml/L) respectively for decellularization of endotheliocytes and fibroblasts. Tissue samples were then processed with HE, trinitrophenal-sirius red staining and elastic fiber staining , observed under scanning electron microscopy and tested their genome DNA. RESULTS The endothelial cells and fibroblasts in experimental group valves and vessel walls were completely decellularizated, no cell fragments were retained within the matrix scaffold; collagen fiber and elastin fiber had been preserved with intact structure and wavily arrayed; DNA content of experimental group valves and vessel walls were decreased by 97.58% ,97.25% compared with that of control group. CONCLUSION The sodium deoxycholate plus Triton-X-100 procedure is an effective decellular technique for bovine jugular venous valved conduits.
