摘要
采用基因组原位杂交(GISH)检测和染色体组成分析方法,对大穗多花小麦新种质4844后代的15个株系进行遗传分析。结果发现,4844-12是1个稳定的异附加系,4844-2和4844-8是稳定的异代换系;对异代换系进行SSR分析表明,代换系中小麦的6D染色体被1对P染色体代换,说明这对冰草染色体与小麦6D染色体有部分同源关系,由此确定4844中的冰草染色体为6P;同时筛选出冰草6P染色体的4个SSR标记。
GISH and SSR were adopted to genetically analyze 15 offspring plants (lines) of the new wheat germplasm 4844 with large and muhiflorous spikes. It was found that 4844-12 was a stable alien additional line and 4844-2 and 4844-8 were stable alien substitution lines;the SSR analysis of the alien substitution lines revealed that in the substitution lines chromosome 6D of wheat were substituted by a pair of P chromosomes and this indicated that there existed a partially homologous relation between the chromosomes of Agropyron cristatum and wheat,and this consequentially confirmed that in 4844 the chromosome from A. cristatum was chromosome 6P. In addition, four SSR markers of chromosome 6P of A. cristatum were screened.
