摘要
用超声法提取丹参饮片中的脂溶性有效成分,然后用反相高效液相色谱法同时测定丹参饮片中隐丹参酮、丹参酮Ⅰ和丹参酮ⅡA的含量。采用DiamonsilTM(钻石)C18色谱柱(250mm×4.6mm,5μm),以V甲醇∶V水=75∶25为流动相,流速为0.8mL/min,检测波长为270nm。3个活性成分隐丹参酮、丹参酮Ⅰ和丹参酮ⅡA分别在1.456~10.19、2.160~15.12和3.152~22.06μg/mL含量范围内与峰面积呈良好线性关系,相关系数r分别为0.9991、0.9992和0.9996;平均回收率隐丹参酮为98.04%(RSD=1.3%,n=3),丹参酮Ⅰ为99.65%(RSD=1.2%,n=3),丹参酮ⅡA为99.31%(RSD=1.2%,n=3)。所建立的质控方法快速、简便、准确,可用于丹参饮片的质量控制。
To use the method of ultrasonic extraction to extract the fat-soluble components in radix salviae miltiorrhizae, the determination of cryptotanshinone, tanshinone Ⅰ and tanshinone Ⅱ A by RP-HPLC. The column was Diamonsil^TM C18 (250×4.6mm, 5μm); the mobile phase was methanol-water(75 : 25), the flow rate was 0.8mL/min, the detection wavelength was 270nm. The calibration curves of cryptotanshinone, tanshinone Ⅰ, tanshinone ⅡA were linear in the range of 1.456~10.19, 2.160~15.12 and 3.152~22.06μg/mL with the correlation coefficient beyond 0.9991, 0.9992 and 0.9996 respectively. The mean recoveries of the assay were 98.04% (RSD = 1.3%, n = 3) for cryptotanshinone, 99.65% (RSD = 1.2%, n = 3) for tanshinone Ⅰ, and 99.31% (RSD = 1.2 %, n = 3) for tanshinone Ⅱ A respectively. The method of quantitative control was rapid, accurcy and reliability, and can be used to control the quality of radix salviae miltiorrhizae.
