摘要
目的:建立高效液相色谱法测定中药复方龙苓春合剂提取物灌胃后大鼠血浆中五味子醇甲的含量,并研究其在大鼠血浆中的药动学行为。方法:采用 Diamonsil^(TM) C_(18)(200mm×4.6mm,5μm)色谱柱,甲醇-乙腈-水(35:35:60,v/v)为流动相,流速:1.0mL·min^(-1),柱温:35℃,检测波长:250nm。以17β-雌二醇为内标,乙酸乙酯萃取后,测定大鼠血浆中五味子醇甲的含量及其药动学参数。结果:大鼠血浆样品中,五味子醇甲的线性范围为0.0128~2.55μg·mL^(-1)(r=0.9991),提取回收率66.71%~76.37%,日内、日间精密度 RSD 均<10%,检测限为4.25ng·mL^(-1)。K_e(h^(-1))为0.27±0.09,t_(1/2)(h)为2.97±1.47,T_(max)(h)为6.33±0.82,C_(max)(μg·mL^(-1))为1.53±0.46,AUC_(0→t)(μg·h·mL^(-1))为14.67±4.61,AUC_(0→∞)(μg·h·mL^(-1))为14.88±4.56。结论:本方法专属准确,中药复方龙苓春合剂的药动学研究结果,为临床用药提供参数依据。
Objective : To develop an RP - HPLC method for determination of schizandrin in rat plasma after ig. the extract of Longlingchun mixture and study pharmacokinetics of schizandrin in rat plasma. Methods:A DiamonsilTM C18 (200 mm ×4. 6 mm, 5 μm)column was used, the mobile phase consisted of methanol, acetonitrile and water (35: 35: 60,v/v) ,the flow rate was 1.0 mL·min^-1 ,the column temperature was controlled at 35 ℃ and the UV detector was operated at 250 nm. 17β - estradiol was used as internal standard when measuring the content and pharmacokinetic parameters of schizandrin in rat plasma. Results:In rat plasma, a good linearity between peak area ratio of schizandrin to the internal standard and the concentration was found in the range of 0. 0128 - 2. 55 μg·mL^-1 (r = 0. 9991 ) ; the extraction recoveries were 66. 71% - 76. 37 % ; within - day RSD and inter - day RSD were both less than 10 %. The limit of test was 4. 25 ng·mL^-1. The main pharmacokinetic parameters was as follows : Ke (h^-1 ) 0. 27 ± 0. 09, t1/2 ( h ) 2. 97 ± 1.47, Tmax ( h ) 6. 33 ± 0. 82, Cmax= ( μg·mL^-1 ) 1.53 ± 0. 46, AUC0→t ( μg·h·mL^-1 ) 14.67 ±4.61 ,AUC0→∞ (μg·h·mL^-1 ) 14.88 ±4. 56. Conclusion:The method is accurate with good reproducibilities. It can be used for the pharmacokinetic studies of Longlingchun mixture.
