摘要
目的:探讨补肾生血药对缺血后肢大鼠外周循环中骨髓来源内皮祖细胞(EPC)的影响及缺血组织血管新生的影响。方法:建立大鼠后肢缺血模型,喂食补肾生血药,1周后细胞培养观察外周血EPC数量变化,4周后检测缺血组织微血管密度及血管内皮生长因子(VEGF)的表达情况。结果:实验组大鼠外周血培养的EPC(89.1±10.3/mm2)明显高于对照组(48.6±7.5/mm2)(P<0.01),缺血组织微血管密度(288.76±12.54)和VEGF表达(0.233±0.014)均高于对照组(224.84±5.87),(0.162±0.018,P<0.05),且外周血EPC数量与微血管密度之间具有显著相关性(P<0.01)。结论:补肾生血药通过动员骨髓EPC的迁移、分化、促进局部的血管新生,达到改善供血目的。
Object:To explore the effect of limb isohemia on mobilization of bone marrow derived-endothelial pregenitor cells(EPCs) and neovasoularization of ischemic tissue with traditional medicine. Methods :Unilateral ischmic model of the hindllmbs was produced by excision of the common and superfioial femoral artery in 20 male rats. The other 20 animals served as the control group. At 1 week after operation the changing of the ciroulating EPCs was observed by mononuolear oells oulture. Miorovesseldensity, the VEGF expression and ansiosraph of isohemlo limbs was detected at week 4. Results:The number of ciroulating EPCs in experiment group (89.1±10.3/mm^2 VS 48.6±7.5/mm^2, P〈0. 01). Mioroveseel density of Isohemio limbs was significoantly higher than that in control group (288.76±12.54/mm^2 VS 224.86±5.87/mm^2 , P〈 0. 01). The VEGF expression in experiment group was signifioantly higer than that in control group (A:0. 233±0.014 VS 0. 162±0. 0186, P〈0.01). The number of circulating EPCs was greatly related to the miorovessel density and VEGF expression (P〈0. 01). Conclusion:Local perfueion of ischemic limb oan be augmented by improving neovasoularization with EPCs mobilization of bushenshensxue traditional medioine.
