摘要
采用不同比例的丙酮、乙醇混合试剂,直接浸泡提取猕猴桃叶中叶绿素,运用Arnon法测定叶绿素含量。分别与丙酮研磨法进行比较,结果表明,丙酮与乙醇体积比为2∶1时,与丙酮研磨法极为接近,提取更为充分。然后选取最佳提取液,在温度、时间、液料比三因素条件下,进行U7(73)实验。结果表明,在35℃,浸泡14h,液料比=12∶1时,叶绿素提取效果最佳,叶绿素得率为2.273mg/g。在最佳方案确定时,进行了叶绿素的分离实验,叶绿素粗品得率为0.184%。
Different rates of acetone and ethanol mixture directly soaking chorophll which extracted from kiwi fruit leaves. An Arnon method was used to determine the content of chorophll. Compared with acetone method, the result showed that the extracting liquid mixed 2 volume of acetone with 1 volume of ethanol was the best method, and chorophll could be extracted completely. In factors of temperature, time, proportion of mixture to the quantity of kiwi fruit leaves, uniformity experiment U7 (7^3) was used to analysis the relationship between the effects. And the optimal conditions were:35℃, extracting 14 hours, the proportion of the mixture to the quantity of leaves 12:1, and the obtaining ration of chorophll is 2.273mg/g. And then, the experiment of isolating chorophll was done, and the obtaining ratio is 0.184%.
