摘要
以陇东马牙枣盛花后40～50 d胚乳为外植体,采用胚剥离和不剥离两种培养方式,进行离体培养．结果表明: 愈伤组织诱导以Nitsch+ZT1．0 mg／L+2．4-D0．6 mg／L+LH0．5 g／L+蔗糖30 g／L培养基最佳,幼胚对胚乳愈伤组织的诱导有促进作用;诱导可获得两类不同的愈伤组织,经染色观察发现两类愈伤组织的细胞形态、色泽、质地、分生能力存在差异．
Endosperms of Zizyphus jujuba cv. Longdongmaya were taken as explants and cultured with different auxin and cytokinin concentration combination after 40～50 days florescence. The immature empryoes were taken from endosperms and not for culturing. The results showed that the Nitsch medium added with 2,4-D 0.6 mg/L＋ZT 1.0 mg/L＋LH0.5 mg/L＋sugar 30 g/L was the best for inducing calli, the immature empryoes were suitable for inducing calli, and two different kinds of calli were got. Through observing the dyed calli, the cell morphology, quality, color and the fission ability between two kinds of calli were different.
