鞘内注入地西泮对足底注射甲醛大鼠躯体痛的抑制效应

Inhibitive effect of intrathecal diazepam on somatic pain induced by plantar subcutaneous injection of formaldehyde in rats

摘要 目的:观察鞘内注入地西泮对大鼠持续性躯体痛的影响。方法:实验于2005-05/07在解放军第四军医大学基础部教学实验中心实验室完成。48只SD大鼠用计算机编号随机分组的方法,分为6组:足底对照组:足底注射0.1mL生理盐水;模型组:足底注射0.1mL20g/L甲醛;鞘内对照组:鞘内给予10μL生理盐水,10min后足底注射0.1mL20g/L甲醛;低剂量实验组、中剂量实验组、高剂量实验组:鞘内分别给予3种不同浓度的地西泮(0.4g/L;1.6g/L;3.0g/L),10min后足底注射0.1mL20g/L甲醛,观察地西泮对大鼠足底皮下注射甲醛引发的持续性躯体痛相关行为(缩足与舔足)的影响。疼痛程度以各组大鼠足底给药后60min内,每5min内以及60min内大鼠缩足与舔足的时间为指标。结果:48只大鼠均进入结果分析。模型组每5min的缩足与舔足时间明显高于足底对照组和实验组(P<0.001);低、中、高剂量实验组足底给药后60min内缩足与舔足总时间比模型组分别减少了57.81%,71.37%,49.45%(P<0.001)。结论:实验结果显示鞘内注射地西泮可以抑制大鼠足底注射甲醛引起的持续性躯体痛,其数据无临床应用意义。 AIM: To investigate the possible effects of intrathecal diazepam on persistent somatic pain in rats. METHODS The experiment was performed at Laboratory of Teaching Experimental Center, School of Basic Medicine, Fourth Military Medical University of Chinese PLA from May to July 2005. Totally 48 SD rats were randomly assigned into 6 groups. The rats in planta control group were injected with 0.1 mL saline in planta. The rats in the model group were injected with 0.1 mL 20 g/L formaldehyde in planta. The rats in the intrathecal control group were given with 10 μL saline, and 10 minutes later 0.1 mL 20 g/L formaldehyde were injected into planta. The rats in the low, middle and high dosage group were injected with diazepam of 0.4 g/L; 1.6 g/L; 3.0 g/L, respectively, and 10 minutes later 0.1 mL 20 g/L formaldehyde were injected into planta. Influence of diazepam on related behavior of successive somatic pain (lifting paws and licking paws) induced by formaldehyde by hypodermical injection in planta was observed and durations of lifting paws and licking paws were recorded for 60 minutes and every 5 minute after administration as the, index of persistent somatic pain. RESULTS: Totally 48 rats were involved in the result analysis. The duration of lifting and licking paws in rats of the model group every 5 minutes was significantly higher than that in the planta control group and experimental group (P 〈 0.001). The duration of lifting and licking paws in rats of the low, middle and high dosage group within 60 minutes after administration decreased by 57.81%, 71.37%, 49.45%(P 〈 0.001) as compared with the model group. CONCLUSION: Experimental results show that intrathecal pretreatment with diazepam can suppress persistent somatic pain induced by formaldehyde injection in planta of rats, and these data are insignificant in clinical application.
