摘要
目的研究mdm2和p53基因在大肠癌中的表达,探讨其在大肠癌局部进展中的作用。方法采用流式细胞术(FCM)对经病理证实的63例大肠癌和11例正常对照组结肠标本分别进行mdm2、p53基因蛋白检测,并进行对比分析。结果大肠癌的发生与mdm2高表达、p53低表达有关,mdm2与大肠癌局部发展、浸润、转移有明显相关性,p53起负调控作用。结论mdm2、p53基因异常表达与大肠癌的发生、淋巴转移有明显相关性。mdm2、p53联合检测对评估大肠癌预后有重要的临床价值。
Objective To study the expression of mdm2 and p53 gene and its role in ontogenesis of colorectal cancer. Methods Flow cytometry technique (FCM) was used to detect mdm2, p53 gene protein of large intestine tissue specimen in 63 patients with colorectal cancer confirmed pathologically [ 31 men and 32 women, mean age (56 ±10) years ] and 11 normal persons [6 men and 5 women, mean age (55 ±9) years]. The results were compared through analysis. Results The occurrence of colorectal cancer is related to high expression of mdm2 and low expression of p53 ; mdm2 is correlated with local development, infiltration and metastasis of colorectal cancer, while p53 plays a negative regulating role. Conclusion The abnormal expression of mdm2 and p53 gene has a obvious correlation with occurrence and lymphatic metastasis of colorectal cancer. The detection of mdm2 and p53 gene in combination is of important clinical value to the prognosis of colorectal cancer.
