参乌胶囊抑制阿尔茨海默病模型小鼠脑内β-淀粉样肽生成的机制研究 被引量:10

摘要 目的 观察中药新复方参乌胶囊对β-淀粉样肽前体蛋白(APP)转基因小鼠脑内β-淀粉样肽(A11)代谢及β-和γ-分泌酶的影响。方法 APP转基因小鼠从4~10月龄灌胃给予参乌胶囊。用免疫组织化学法观察APP、Aβ、β位点剪切酶(BACE)和早老蛋白1(PS-1)的表达,并进行图像分析。结果 10月龄APP转基因小鼠模型组与同月龄正常对照组相比,脑内海马区APP、Aβ和PS-1(为γ-分泌酶的催化部位)的表达明显增多,而BACE(即β-分泌酶)的变化不明显。参乌胶囊能明显减少APP转基因小鼠脑内海马区APP的表达,减少脑内All的产生,抑制BACE和PS-1的表达。结论 参乌胶囊抑制脑内All的产生可能是通过减少APP含量及抑制β-和1-分泌酶而起作用的。 Objective To investigate the effects of Chinese herb compound Shen-wu capsule on the metabolism of β-amyloid (Aβ) and β- and γ- seeretase in β-amyloid precursor protein (APP) transgenic mice. Methods APP transgenic mice were administered intragastrically by Shen-wu capsule or water from 4 to 10 months old. The expression of APP, Aβ,β-site APP cleaving enzyme (BACE) and presenilin-1 ( PS-1 ) in hippoearnpus CA1 region was detected by immunohistoehemistry with image analysis. Results Compared with normal group, the expression of APP,Aβ and PS-1 ( the catalytic site of β-secretase) increased obviously in the model group of 10-month-old APP transgenic mice;there was no significant changes in the expression of BACE ( i. e. β-secretase). Shen-wu capsule decreased the expression of APP,reduced the production of Aβ, and inhibited the expression of BACE and PS-1 in the hippoearnpus of APP transgenic mice. Conclusions Shen-wu capsule could inhibite the production of Aβ through decreasing the content of APP and inhibiting β- and γ-secretase.
