心脑舒通胶囊对高脂血症合并心肌梗死大鼠心肌细胞膜Na^+-K^+-ATPase、Ca^(2+)-Mg^(2+)-ATPase活性及乳酸含量的影响 被引量:6

Effects of Gross Saponins of Tribulus on Na^+-K^+-ATPase and Ca^(2+)-Mg^(2+)-ATPase Activity and Lactic Acid Content in Hyperlipemic Rats with Acute Myocardial Infarction

摘要 目的观察心脑舒通胶囊(蒺藜总皂苷,GrossTotalSaponinsofTribulus,GSTT)对高脂血症大鼠急性心肌梗死(AcuteMyocardialInfarction,AMI)后心肌细胞能量代谢及乳酸代谢的影响。方法SD大鼠高脂饲料喂养15d,眼眶采血筛选高脂血症大鼠,结扎冠状动脉左前降支,体表心电图Ⅱ导示ST段弓背抬高,证实AMI形成后,将大鼠随机分为5组:正常组(10只);AMI高脂模型组(13只);AMI高脂加GSTT大剂量组(24.3mg·kg-1·d-1,13只);AMI高脂加GSTT小剂量组(12.15mg·kg-1·d-1,13只);AMI高脂加辛伐他汀组(3.6mg·kg-1·d-1,13只)于手术后第2日开始灌胃给药,连续28d。然后截取左室缺血区心肌,制备心肌线粒体,观察GSTT对心肌梗死后大鼠心肌细胞膜Na+-K+-ATPase、Ca2+-Mg2+-ATPase活性及乳酸代谢的影响。结果模型大鼠心肌细胞膜Na+-K+-ATPase、Ca2+-Mg2+-ATPase活性明显降低,而乳酸含量却明显增强;与模型组比较,GSTT可明显提高心肌细胞膜Na+-K+-ATPase、Ca2+-Mg2+-ATPase活性(P<0.05和P<0.01),且能够降低缺血心肌乳酸含量(P<0.05)。结论GSTT可显著改善高脂血症大鼠AMI后心肌的能量代谢及乳酸代谢。 Objective To observe the effects of Gross Saponins of Tribulus terrestris (GSTT) on energy metabolism and lactic acid metabolism in hyperlipemic rats with acute myocardial infarction (AMI). Methods Rats were fed with high fat diet for 15 days to induce hyperlipidemia, then left anterior descending branch of coronary artery of rats were ligated. Rats showed ST segment elevation in lead 11 of electrocardiogram were proved to be AMI rats. AMI rats with hyperlipidemia were divided randomly into five groups (normal group, model group, 24. 3 mg ·kg^-1 GSTT group, 12. 15 mg· kg^-1 GSTT group, Simvastatin group ) and test drugs were given in succession for 28 days. Left ventricular ischemic myocardium were harvested to prepare myocardial mitochondria. Na^+-K^+-ATPase and Ca^2+-Mg^2+-ATPase activity and lactic acid content were measured. Results Compared with the normal group, Na^+-K^+-ATPase and Ca^2+-Mg^2+-ATPase activity significantly lowered in model group ( P 〈 0. 01), while lactic acid content increased significantly ( P 〈 0. 01 ). Compared with the model group, large - dosage of GSTT group, low - dosage of GSTT group and Simvastatin group can improve Na^+-K^+-ATPase and Ca^2+-Mg^2+-ATPase activity ( P 〈 0. 01, P 〈 0.05), lower lactic acid content in ischemic myocardium ( P 〈 0.05). Conclusions GSTT has the effect of improving myocardial energy metabolism and lactic acid metabolism in hyperlipemic rats with AMI.
