摘要
目的:采用反相高效液相色谱-蒸发光散射检测法(HPLC—ELSD)同时测定枸橼酸钾中枸橼酸和钾的含量。方法:采用Synergi Fusion-RP80色谱柱(250mm×4.6mm,4μm),以甲醇-水(含甲酸1%)(5:95)为流动相,流速为1mL/min,柱温为室温;ELSD检测条件:漂移管温度为40℃,雾化气体压力为3.5bar。结果:枸橼酸在0.24~11.85μg范围内,进样量与峰面积呈良好的线性关系,r=0.9992,平均回收率为101.9%(n=9),RSD为2.5%(n=6);钾在0.07~3.61μg范围内,进样量与峰面积呈良好的线性关系,r=0.9994,平均回收率为101.8%(n=9),RSD为1.3%(n:6)。结论:本方法可用于枸橼酸钾中枸橼酸和钾的同时定量测定。
Aim: To simultaneously determine citric acid and potassium in potassium citrate by HPLC-ELSD. Methods: The developed method was based on the separation of citric acid and potassium on Synergi Fusion-RP80 column (250 mm × 4.6 mm, 4 tan) at ambient temperature using methanol-water (containing 1% formic acid) (5:95) as the mobile phase and a flow rate of 1 mL/min; ELSD was used as the detector with nebulizing air pressure of 3.5 bar and the drift tube temperature of 40℃. Results: A linear range of the injected amount of citric acid between 0.24μg and 11.85μg over the responses and the mean recovery of 101.9% (n = 9)with RSD of 2.5% (n = 6)were obtained. And it was also found that a linear range of potassium between 0.07 μg and 3.61 μg with the mean recovery of 101.8% (n = 9)with RSD of 1.28% (n = 6)for potassium existed. Conclusion: A simple and accurate method was established for the simultaneous detemlination of potassium and citric acid in potassium citrate and its sustained-release tablets.
