金雀黄素对子宫内膜癌细胞体外生长影响的研究 被引量:1

摘要 目的研究金雀黄素抑制人子宫内膜癌细胞系Ishikawa细胞体外增殖作用机理。方法人子宫内膜癌细胞系Ishikawa细胞进行体外培养,用MTT法检测细胞增殖作用,用流式细胞仪检测细胞周期情况。结果金雀黄素能明显抑制Ishikawa细胞体外增殖,并且随着药物浓度增加其抑制作用更加明显,呈剂量依赖性,金雀黄素对Ishikawa细胞体外增殖抑制最显著的时间是加药48h,加药后72h的抑制率虽然有所上升,但不甚明显,加药96h其抑制率稍低,加药后120h其抑制率明显降低;金雀黄素处理的Ishikawa细胞明显阻滞于G2-M期。结论金雀黄素通过阻滞细胞于G2-M期,从而抑制Ishikawa细胞体外增殖。 Objective To study the inhibitory mechanism of genistein on extracorporeal proliferation of ishikawa cells of human endometrial cancer cell line. Methods Ishikawa cells of human endometrial cancer cell line were cultured in vitro and then the proliferation of cells by MTT method and cell cycle was detected by flow cytometry. Result Genistein inhibited the proliferation of ishikawa cells in vitro with a characteristic of dose-dependence. The higher of genistein concentration was, the more remarkable of genistein effect was. The inhibitory effect was most obviously 48 hours after the administration of genistein. After 72 hours the inhibitory rate was increased slightly without significance and after 96 hours it was decreased slightly. The inhibitory rate was significantly decreased 120 hours after the administration of genistein Ishikawa cells effected by genistein were blocked in G2-M phase significantly. Conclusion Genistein can inhibit the extracorporeal proliferation of ishikawa cells through blocking the cells in G2-M phase.
