摘要
目的观察电针对局灶性脑缺血再灌注大鼠白细胞介素-6(IL-6)、白细胞介素-10(IL-10)表达的影响,探讨针刺治疗缺血性脑损伤的可能机制。方法采用血管内线栓法制作SD大鼠大脑中动脉梗塞模型(MCAO),梗塞2h后拔出栓线造成再灌注损伤,观察缺血再灌注后不同时间点以及电针治疗后大鼠血浆和脑组织中IL-6、IL-10含量的变化。结果治疗1组外周血IL-6、IL-10比模型对照1组升高(P<0.05),治疗2组IL-6较模型对照2组升高(P<0.05)。脑组织IL-10治疗1组与模型对照1组差异显著(P<0.05)。结论电针可通过在一定范围内增强IL-6、IL-10的表达来抑制炎性细胞在缺血区的聚集和激活,而抑制细胞毒性物质的释放,从而改善微循环,促进大脑血供,有利于大鼠脑神经功能的恢复。
Objective To observe the influence of electroacupuncture on interleukin-6 (IL-6) and interleukin-10 (IL-10) in rats with local cerebral ischemia-reperfusion and investigate the mechanism of acupuncture for treating cerebral ischemia lesion. Method The model of middle cerebral artery obstruction (MCAO) was established in SD rats by using intraluminal suture. After 2 hours the suture was withdrawn to induce reperfusion lesion. The changes of IL-6 and IL-10 contents were observed in rat plasma and cerebral tissue at different time points and after the treatment of electroacupuncture. Result The contents of IL-6 and IL-10 were increased in the treatment group 1 than that in the control group 1 (P〈0.05). The content of IL-6 in the treatment group 2 was increased than that in the control group 2 (P〈0.05). There was a significant difference in IL-10 content between the treatment group 1 and control group 1 (P〈0.05). Conclusion Electroacupuncture can inhibit the aggregation and activation of inflammatory cells and the release of cell toxic materials in ischemia area through improving the expressions of IL-6 and IL-10 to a certain extent. Electroacupuncture can improve the microcirculation, cerebral blood supply and the recovery of rat cranial nerve.
