Taq Man探针定量PCR对癫痫大鼠脑组织GLAST、GLT-1表达变化的研究 被引量:4

Quantitative Detection of Real-time Fluorescent PCR on the Expression of GLAST and GLT-1 in Rats with Epilepsy

摘要 目的探讨谷氨酸转运蛋白GLAST、GLT-1不同亚型在癫痫发作过程中的表达变化。方法采用TaqMan探针实时定量PCR(real-time quantitative PCR)技术,检测癫痫发作后不同时点脑组织中谷氨酸转运蛋白GLAST、GLT-1表达量的变化。结果癫痫发作后不同时点GLAST、GLT-1在大脑中的表达量明显低于对照组(P<0.01),且在大脑中不同时点的表达量具有明显的差异(P<0.01);GLAST在小脑中不同时点的表达量无明显差异(P>0.05),但均明显低于对照组(P<0.01);GLT-1在小脑中无表达。结论癫痫发作后谷氨酸转运蛋白GLAST、GLT-1在大脑中的表达量明显减少;GLAST、GLT-1表达量的减少可能是癫痫发病的诱因之一。 Objective To investigate the expression of GLAST and GLT-1 in rats with epilepsy. Methods Glutamate transporter GLAST and GLT-1 genes expression were detected by real-time fluorescent quantitative PCR assay with TaqMan probe in different time after epileptic seizure. Results The expression amount of GLAST and GLT-1 in cerebrum were obviously lower than that of the control in different time after epileptic seizure (P 〈 0.01). Furthermore, the amount of expression had evidently difference in cerebrum in different time after epileptic seizure (P 〈 0.01). The expression amount of GLAST in cerebellum was not significantly different in different time after epileptic seizure (P 〉 0.05), but they were obviously lower than that in the control (P 〈 0.01). However, GLT-1 was not expressed in cerebellum. Conclusion The expression amount decrease of GLAST and GLT-1 may be one of the factors of epileptic seizure.
