摘要
通过对红肉脐橙成年态茎段进行离体培养,研究了不同条件对茎段培养污染率的影响。结果表明:在炎热干燥的夏季午后取长0.8cm左右半木质化茎段,用浓度为0.1%HgCl2消毒20min,间隔24h后再用浓度为2%NaClO处理15 min之后进行培养,污染率最低,可控制在10%以内。
The experiment in mature citrus stem segment culture in vitro was carried out and contamination rate in different conditions was studied. The results indicated that: the half senescent segments of about 0.8cm which were token from the field at :2 pm in sunny summer day as explants were best. The segments treated with 0.1% HgCl2 for 20 minutes, and 24 h later,:2 % N aClO for 15 minutes, could keep the contamination rate of the citrus stem segment culture below 10 %.
