摘要
提取了7个药用菟丝子种样品的DNA,采用ITS序列扩增通用引物对其进行PCR扩增,并对PCR产物进行克隆和测序.根据所测得菟丝子ITS全长序列与其常见易混品所在属的代表植物ITS序列比对,设计了一对特异性引物并进行了PCR检测.结果表明,此对引物能将药用菟丝子种子与4种易混品区分开来,可以用于药用菟丝子的真伪鉴定.
DNAs of 9 sets of samples from 7 officinal Cuscuta spp. were extracted.The ITS sequences by comparing the obtained sequences with the ITS sequences of 4 fake plant spieces in other family, the specific PCR primers were designed and in further PCR detection proved effective in distinguishing Cuscuta sp.from the others. Finally it is concluded that this method can be in practice used to identify officinal Cuscuta.
