摘要
用十六烷基三甲基溴化铵(CTAB)沉淀法从透明质酸(HA)发酵液中提取透明质酸。考察了该方法中稀释倍数、pH、盐浓度和CTAB的体积分数等因素对HA提取效率的影响。结果表明,在将发酵液稀释至4.5倍、调节pH=6.5和加入发酵液体积的12%的CTAB溶液的条件下,HA收率可高达97.0%。与乙醇法所得透明质酸沉淀的纯度和收率相比,CTAB法得到的透明质酸的收率提高了2.1%,杂蛋白降低了2.4%。与氯代十六烷基三甲基吡啶法相比,CTAB法的成本比CPC法的少45.2%。
The method with CTAB were developed to separate HA from fermentation broth. The optimal conditions of HA recovery with CTAB were determined. The fermentation was diluted by distilled water, and the volume ratio of the broth to water is 1:3.5(v/v). The pH of the culture diluted was adjusted to 6.5. Then, 10 % ( w/v ) CTAB was added into the culture diluted to finanal concentration 12 % ( v/ v ). The yield was up to 97.0 %. The methods with CTAB and with alcohol to separate HA from fermentation broth was compared. Of the two the purity and the yield of the latter were better than the former. The cost is compared between the methods with CPC and with CTAB, the latter is lower.
