螺内酯对大鼠肾间质纤维化的保护作用

摘要 目的观察螺内酯对单侧输尿管梗阻(UUO)大鼠的肾皮质纤溶酶原激活物抑制物-1(PAI-1)表达介导的肾间质纤维化的作用。方法UUO大鼠给予螺内酯20mg.kg-1.d-1灌胃,用RT-PCR方法检测术后7、14、21dPAI-1mRNA的表达。结果与假手术组相比,UUO组及药物治疗组的PAI-1mRNA表达水平均增高(P<0.05),但治疗组低于UUO组(P<0.05)。结论螺内酯可通过下调PAI-1减轻UUO术后肾组织间质纤维化。 Objective To investigate the effect of aldosterone antagonist (spironolactone) on the expression of type 1 plasminogen activator inhibitor (PAI - 1 ) in renal interstitial fibrosis following unilateral ureteral obstruction (UUO) in rat kidney. Methods UUO group, spironolactone (20 mg·kg^-1·d^-1) was given, RT -PCR was performed to investigate the expression of PAI- 1 mRNA on the 7, 14 and 21 day in the kidney. Results In comparison with the sham group, the expression of PAI- 1 mRNA of UUO and medication groups increased significantly ( P 〈 0, 05 ), meanwhile spironolactone could significantly decrease the expression of PAI- 1 as compared to UUO group ( P 〈 0. 05 ). Conclusion Spironolaetone may play a protective role in renal interstitial fibrosis following UUO through the down - regulation of PAI - 1.
