摘要
目的观察葛根素对实验性脑缺血谷氨酸脱羧酶(GAD67)表达变化的影响。方法应用原位杂交方法检测大鼠永久大脑中动脉闭塞(MCAO)模型GAD67mRNA在脑缺血后不同时间的表达。结果正常组及假手术组GAD67mRNA在脑组织中表达较弱。对照组和治疗组GAD67mRNA在脑缺血后6h即开始表达增强,在72h 达高峰,后逐渐下降,GAD67mRNA表达至第2周仍高于正常,治疗组GAD67mRNA表达高于对照组。结论葛根素可以促进实验性脑缺血后GAD67mRNA的表达,具有神经保护作用,有助于神经功能的恢复。
Objective To study the expression of GAD67mRNA after experimental cerebral ischemic damage in rats and explore the neuroproteetive role of puerarin. Methods 84 healthy Wistar rats were recruited to estab lish the middle cerebral artery occlusion (MCAO) model by the ligation of the right carotid artery. The focal is chemic tissue was fixed ,dehydrated ,and paraffin-embedded in regular way at the time 6h,1d,3d,Sd,Td and 14d af ter the occlusion of MCA respectively. The gene expression of GAD in focal ischemic tissue of different times was assayed by in situ hybridization (ISH). Results The results of ISH revealed that the expression of (;AI)67mRNA was not so high as in the common brain tissue. The gene expression of GAD67 in the controlled and treatment group increased from 6h after the MCAO,and reached their peaks at the time of 72h after ischemia,then declined gradually. The gene expression of GAD67 was higher than that of normal group after 2w. The gene expression of GAD67 in the treatment group was more than that of controlled group. Conclusion Puerarin can promote the gene expression of GAD67. It is neuroprotective and is good to the rehibition of the neurological function.
