摘要
目的:探讨桃核承气汤对大鼠肠缺血再灌注损伤(I/R)的保护作用及其机制。方法:采用肠系膜上动脉夹闭方法制作肠缺血再灌注损伤模型。实验大鼠随机分为正常组、假手术组、缺血再灌注组、桃核承气汤大剂量组、桃核承气汤中剂量组、桃核承气汤小剂量组、乳果糖组。观察肠缺血1 h再灌注3 h后肠黏膜损伤程度,肠组织含水量,测定血浆D-乳酸和内毒素水平,并测定血清IL-6、TNFα-含量。结果:桃核承气汤大、中、小剂量组及乳果糖组肠黏膜损伤程度轻,肠组织含水量低,D-乳酸、内毒素、IL-6、TNF-α均低于缺血再灌注组。结论:桃核承气汤通过减轻炎症反应对大鼠肠缺血再灌注损伤有保护作用,并呈剂量依赖效应。
Objective: To assess the protective effects and explore the mechanism of Taohe Chengqi decoction on intestinal ischemia-reperfusion(IIR) injury in rats. Methods:56 adult Wistar rats(male and female in half) were randomly divided into normal group, sham operated group, IIR group, pretreatment with Taohe Chengqi decoction (large/middle/small dose) groups and lactulase control group. In the experiments, the root of superior mesenteric artery was blocked with non-injury artery clap for 1 hour and reperfusion for 3 hours. Then observed the damage of intestinal mucous by histopathology, detected the amount of water content in rat's intestine tissue, measured the level of plasma D-lactate and endotoxin, and the content of tumor necrosis factor-alpha(TNF-α) and interleukin-6(IL-6) in serum. Results:Compared with the IIR group, the damage of intestinal mucous score , the amount of water content, the level of D-lactate, endotoxin, TNF-α and IL-6 in Taohe Chengqi decoction and lactulose groups were obviously decreased, Conclusion: Taohe Chengqi decoction can protect rats from intestinal ischemia-reperfusion injury by suppressing inflammatory response, and the effect is exerted in a dose-dependent manner.
