摘要
天冬氨酸特异性半胱氨酸蛋白酶(C aspase)是与细胞凋亡有关的一类蛋白酶,其中C aspase-3是细胞凋亡的最终执行者。为了进一步研究C aspase-3的生物学功能,从PK-15细胞中提取总RNA,用RT-PCR方法扩增出猪C aspase-3基因,序列分析表明克隆的C aspase-3基因与G enB ank上登录的猪C aspase-3基因核苷酸与推导的氨基酸序列同源性分别为98.9%和98.6%,与人和鼠的C aspase-3基因核苷酸、氨基酸序列同源性分别为88.6%、83.5%和87.8%、86.7%,而且猪、人、鼠的C aspase-3上的催化和剪切位点都极为保守。
Caspase is a kind of proteases which is related to apoptosis, and Caspase-3 is the ultimate execute in apoptosis. In order to study the biological function of Caspase-3, the Caspase-3 gene of pig was amplified by RT- PCR with the extraction of RNA from PK-15 cells. After cloning and sequencing, the results showed that the nucleotide and the deduced amino acid sequences of cloned Caspase-3 shared 98.90% and 98.60% homology with those of Caspase-3 published in GenBank. As compared with that of human and mouse, the Caspase-3 gene homology of the nucleotide and the deduced amino acid sequences were 88.6%, 83.5% and 87.8%, 86.7%, respectively. The active sites corresponding catalysis and cleavage on Caspase-3 among the pig, human and mouse were conservative very much.
