人参二醇组皂苷及人参皂苷Rb_1对体外培养黑色素细胞的影响 被引量:10

The Effects of Panaxadiol Saponins and Ginsenoside Rb_1 on Cultivation Melanocytes in vitro

摘要 目的探讨人参二醇组皂苷、人参皂苷Rb1对体外培养的黑色素细胞及黑色素合成的调节作用及机制。方法①将黑色素细胞进行体外培养、传代;用人参二醇组皂苷、人参皂苷Rb1进行干扰,观察细胞的生长情况,测量细胞OD值;②免疫组织化学技术显示黑色素细胞的一氧化氮合酶的表达的改变。结果对照组与人参二醇组皂苷实验组、人参皂苷Rb1实验组的细胞生长OD值无显著性差异(P>0.05),对照组与人参二醇组皂苷实验组、人参皂苷Rb1实验组的一氧化氮合酶表达有显著性差异(P<0.05)。结论人参二醇组皂苷、人参皂苷Rb1对黑色素细胞的生长、增殖无影响,但对培养黑色素细胞一氧化氮合酶的表达具有抑制作用。 Objective To discuss the effects and regulation mechanism of Panaxadiol saponins and Ginsenoside Rb1 on cultivation melanocytes and melanin synthesis in vitro. Methods (1)Culture and passage the melanocytes in vitro ; interfere the melanocytes with Panaxadiol saponins and Ginsenoside Rb1 ,then observe the growth of the cells and measure the OD;(2)Display the change of expression of nitric oxide synthase in the melanocytes with immunohistochemistry. Results The OD had no significant difference between the control group and treatment group of Panaxadiol saponins or Ginsenoside Rb1 (P 〉 0.05 ), the expression of nitric oxide synthase had significant difference between the control group and treatment group of Panaxadiol saponins or Ginsenoside Rb1 (P 〈 0.05 ). Conclusion The Panaxadiol saponins and Ginsenoside Rb1 can not affect the growth and proliferation of melanocytes, but can inhibit the expression of nitric oxide synthase in melanocytes.
