目的:观察18β-甘草次酸钠(β-SG)对体外培养的小鼠腹腔巨噬细胞摄取氧化型低密度脂蛋白(oxLDL)的影响,并探讨其可能的作用机制。方法:收集并培养小鼠腹腔巨噬细胞,分为对照组、oxLDL组及oxLDL+β-SG(1、10、100μmol/L)组,经oxLDL(除对照组外)作用后测定细胞内总胆固醇(TC)、游离胆固醇(FC)、胆固醇酯(CE)含量,观察不同浓度β-SG对巨噬细胞摄取oxLDL的影响;同时,测定细胞内丙二醛(MDA)的含量和超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-PX)的活性。结果:oxLDL能显著增加巨噬细胞内TC、CE的含量,而预先加入β-SG的oxLDL组能剂量依赖性地抑制细胞内TC、CE含量的升高(P<0.01或P<0.05),FC各组无显著性差异(P>0.05);与oxLDL组比较,oxLDL+β-SG各组还能减少细胞内MDA含量,增加SOD和GSH-PX活性(P<0.01或P<0.05)。结论:β-SG能显著抑制体外培养的小鼠腹腔巨噬细胞对oxLDL的摄取,此作用可能与其能提高巨噬细胞的抗氧化能力有关。
OBJECTIVE: To study the effect of 18β-sodium glycyrrhetate (β-SG) on the uptake of oxidized low density lipoprotein (oxLDL) in peritoneal macrophages of mice cultured in vitro and to study its possible mechanism. METHODS:Peripheral macrophages of mice were cultured and collected, then divided into control group, oxLDL group and oxLDL plus β-SG(1, 10 and lOOμmol/L)group. The contents of total cholesterol (TC), free cholesterol (FC) in macrophages after being treated with oxLDL(with the exception of the control group) were determined; and the effects of β-SG at different concentrations on macrophages' intake of oxLDL were monitored, meanwhile the level of malondialdehyde (MDA) and the activities of superoxide dismutase (SOD) and glutathione peroxidase (GSH- PX) in macrophages were determined. RESULTS: The contents of TC and CE in macrophages were increased significantly by oxLDL, however, which were inhibited dose-dependently in macrophages in oxLDL group pre- treated with β SG (P 〈 0.01 or P 〈 0.05). There was no significant difference among FC groups(P 〉0.05). As compared with oxLDL group, oxLDL plus β - SG group decreased MDA content in macrophages and increased the activities of SOD and GSH- PX (P 〈 0.01 or P 〈 0.05). CONCLUSION: β-SG can significantly inhibit mice peripheral macrophages' uptake of oxLDL, this might be associated to its action in enhancing of peripheral macrophages' antioxidation.
