肾消康浓缩丸对糖尿病骨质疏松大鼠骨形态学影响的实验研究 被引量:9

摘要 目的:通过建立糖尿病合并骨质疏松模型,制备糖尿病合并骨质疏松大鼠胫骨的光镜和电镜标本,观察肾消康浓缩丸对改善糖尿病大鼠骨质疏松状态的疗效。方法:W istar雄性大鼠73只,随机选15只为空白对照组,以普通饲料喂养,经左下腹注射柠檬酸缓冲液25mg/kg。其余58只称重,以25mg/kg链脲佐菌素(STZ)左下腹腔注射,造模成功后再随机分为模型组、治疗组,两组均喂以高热量饲料13W。治疗组以肾消康浓缩丸灌胃1次/天;空白组、模型组以生理盐水灌胃1次/天。26W末处死大鼠取胫骨制备光、电镜标本,并对其进行形态学观察。结果:模型组大鼠胫骨骺板变薄,骨小梁裂纹、断裂者较多,网状结构破坏。治疗组与模型组相比骺板增厚,骨小梁排列及走行基本达到正常组水平,立体网状结构得到一定程度修复。结论:肾消康浓缩丸可有效地增加骺板厚度,促进骨小梁再生,改善骨小梁细薄脆弱的病理状态。 Objective: In order to study the effects of Shen - Xiaokang in the treatment of osteoporosis, a model of diabetic osteoporosis was made in Wistar laboratory rats. An experiment was then conducted to study the effect of the Chinese medical treatment on the tibia of the animals. Tibia samples were obtained and prepared in order to study the bone structure with optical microscopy and electron microscopy. Method: Seventy - three male Wistar rats were randomized: Fifteen were assigned to the control group, fed with normal animal feed and injected with sour buffered lemon extract 25mg/kg. The remaining fifty - eight were weighed, and injected with STZ to build the diabetic model. These fifty - eight were then randomly divided into a model group and a treatment group and were fed with high calorie feed for thirteen weeks. The treatment group rats were administered Shen - Xiaokang once a day while the control group and the model group were given physiological saline once a day. At the end of the twenty - six weeks, the rats were then killed and samples were obtained to observe a sample of the bone structure. Result: The model group rats bone epiphysis were thinner, the trabecular cracks were more numerous and the net - structure was destroyed. While the treatment group rats bone epiphysis was thicker, the trabecular pattern was similar to the normal group. The net - structure was repaired to a certain degree when compared with the model group. Conclusion: Shen - Xiaokang can increase the thickness of bone epiphysis available, can stimulate trabecular regeneration, as well as improve the general condition of osteoporosis.
