摘要
碱性成纤维细胞生长因子(bFGF)可诱发血管形成、刺激癌细胞生长。本研究试图通过检测bFGF及其受体FGFR-1在正常肾和肾癌组织的表达方式,探讨其临床意义。作者针对36例石蜡包埋的肾癌及其正常肾组织,配合微波炉抗原修复法进行bFGF和FGFR-1的LSAB免疫组化染色。结果显示:bFGF与FGFR-1的染色结果基本一致;全部正常肾组织和29例肾癌的细胞外基质为非均一性阳性染色;12例原发性肾癌、2例癌转移灶和体外培养的GRC-1细胞的癌细胞胞浆为阳性染色,且多为均一性染色。癌细胞胞浆的bFGF染色多见于T3-4期肿瘤,与T1-2期比较有显著性差异(P=0.02)。作者认为,bFGF和FGFR-1可同时表达于正常肾和肾癌组织,提示bFGF和FGFR-1的表达与功能密切相关;癌细胞胞浆bFGF阳性表达与肿瘤分期有关,提示bFGF是促进肾癌发展的原因之一。
Basicfibroblastgrowthfactor(bFGF)isapluripotentpolypeptidewhichplaysanim-portantroleintumorprogresionandangiogensis.Wedeterminedtheexpressionandlocalizationpat-ternsofbFGFandoneofitsreceptor(FGFR-1)innomalrenalaswelasinrenalcancers.Theresultswerecomparedwithclinicopathologicfeatures.UsingbFGFandFGFR-1antiboty,therepairingmethodofantigenwithmicrowaveovenheatingandLSABimmunohistochemistryweusedin36casesofparaf-finembeddedrenalcelcarcinoma(RCC)andtheirpairednormalrenaltissues.TheexpressionofbFGFandFGFR-1wasnearlyconsistent.ThenormalrenaltissuesandECMof29casesofrenalcancertisuesshowedheterogenousimmunoreactivities.Renalcancercelcytoplasmof12primarytumorsand2metastatictumors,asinthecytoplasmicbFGFofculturedGRC-1cels,werepositivelyhomogenousstained.ThebFGFandFGFR-1canbeconsistentlyexpressedinnormalrenalandrenalcancertissues,reflectingthattheexpressionandfunctionofthesesubstanceswerecloselyasociated.ThecytoplasmicbFGFexpressionofrenalcancerwasrelatedtotumorstages,suggestingthatbFGFplaysanimportantroleintheprogressionofrenalcelcarcinoma.
