猴头菌转化银杏叶提取物(EGB)条件的研究 被引量:5

Optimization of the Culture Conditions of the Biotransformation of Ginkgo biloba L. Extract(EGB) by Hericium Erinaceus

摘要 以抑制非酶糖基化反应为指标,研究了猴头菌转化银杏叶提取物的培养基组成和培养条件对其活性的影响。研究表明,葡萄糖是最适合的碳源,发酵液抑制率达到94.5%;氮源以蛋白胨效果最好,其抑制率达94.5%;以麸皮和玉米粉为复合营养源的猴头发酵液同样提高了抑制率。进一步进行正交实验,得到猴头菌转化银杏叶提取物培养基的较佳组成为:葡萄糖2%、蛋白胨0.4%、麸皮2%,玉米粉1.5%,EGB 0.5%,KH2PO40.012%,MgSO40.006%,CaCO31%。在此条件下,获得最大抑制非酶糖基化活性为98.1%,比优化前得到显著提高。 Effects of carbon sources, nitrogen sources and growth factors on the bioactivity inhibition of nonenzymatic glycation reaction of biotransformation products of Ginkgo biloba L. extract (EGB) by Hericium erinaceus were investigated by one-factor-at-a-time experiment. The results showed that carbon sources, nitrogen sources and growth factors had significant influence on bioactivity of the biotransfonnation products. By means of the orthogonal experiment design, the optimum media composition for biotransformation of Ginkgo biloba L. extract (EGB) by Hericiurn erinaceus in shaking flask culture was: glucose 2 %, peptone 0.4 %, wheat bran 2 %, corn flour 1.5 %, EGB 0.5 %, KH2PO4 0.012 %, MgSO4 0.006 %, CaCO3 1%. Under above conditions, the highest inhibition ratio of the biotransformation products of Ginkgo biloba L. extract (EGB) by Hericiurn erinaceus was achieved at the level 98.1%.
