蒙古马肌生成抑制基因(MSTN)的克隆及序列分析 被引量:5

摘要 肌生成抑制基因(MSTN)在发育和成熟的骨骼肌中特异表达,并对肌肉发育和再生具有负调控作用。依据马肌生成抑制素(MSTN)编码序列设计引物,以蒙古马基因组DNA为模板,利用PCR技术,扩增出MSTN基因的几个片段后,测序并按顺序拼接,得到DNA全序列。序列分析结果表明,MSTN基因由2个内含子和3个外显子组成,碱基数量分别为第一外显子373 bp、第二外显子374 bp、第三外显子381 bp;第一内含子1 833 bp、第二内含子2 018 bp,共有4 979个碱基。该序列首次登录到GenBank,登录号为AY840554。 Myostatin(MSTN) was expecially expressed on skeletal muscle and was negalitively regulated to development and regeneration of muscle. The complete sequence of Mongolian horse MSTN gene were firstly amplified by PCR technique . The sequence analysis showed that the MSTN gene was composed of two introns and three exons. The sequence lengthes of the first exon , the second exon and the third exon were 373 bp , 374 bp and 381 bp respectively. The lengthes of the first intron and the second intron were 1 833 bp and 2 018 bp respectively. The full length of Mongolian horse MSTN gene was 4 979 bp and firstly submitted to Genbank. Its accession number was AY840554.
