摘要
利用CTAB-异硫氰酸胍法、改良CTAB法和RNAplant试剂盒法,从富含多糖和酚类物质的苹果组培苗继代苗中提取总RNA。结果表明CTAB.异硫氰酸胍法对苹果组培苗总RNA的提取有较好的效果,所得RNA的A260/A瑚大于2.0,A260/‰为1.8—2.0,产率在16.95μg/g左右。改良CTAB法和RNAplant试剂盒法提取的RNA均有少量DNA污染,但RNAplant试剂盒法比CTAB法更快捷,简便。
The CTAB-guanidinium method, modified CTAB method, and RNAplant Kit method were used to extract the total RNA from the plantlets of apple cultured in vitro, in which contain a lot of carbohydrates and phenolic compound. The CTAB-guanidinium method showed the best results in term of the quality and quantity of yielded RNA with the A260/A230 value of more than 2.0, A260/A2s0 ranged from 1.8 to 2.0, and the total RNA yield of 16.95μg/g. The modified CTAB and RNAplant Kit methods were a bit short in total RNA extraction because of a little DNA contamination. And the RNAplant Kit method showed advantages of fastness and convenience.
